The trophoblast clock controls transport across placenta in mice

چکیده
ABSTRACT In mammals, 24-h rhythms of physiology and behavior are organized by a body-wide network clock genes proteins. Despite the well-known function adult circadian system, roles maternal, fetal placental clocks during pregnancy poorly defined. mature mouse placenta, labyrinth zone (LZ) is origin key for selective nutrient waste exchange. Recently, gene expression has been detected in LZ other tissues; however, there no evidence controlled clock. Here, we demonstrate that specifically trophoblast layer harbors an already functional late gestation, able to regulate manner activity xenobiotic efflux pump, ATP-binding cassette sub-family B member 1 (ABCB1), likely gating exposure drugs from maternal circulation certain times day. As more than 300 endogenous exogenous compounds substrates ABCB1, our results might have implications choosing treatment time when aiming either maximal/minimal drug availability fetus/mother.

Nutrient transport across the placenta.
The placenta forms a selective barrier that functions to transport nutrients that are of critical use to the fetus. Nutrient transport across the placenta is regulated by many different active transporters found on the surface of both maternal and fetal facing membranes of the placenta. The presence of these transporters in the placenta has been implicated in the facilitation of nutrient diffus...
متن کاملMediation of Folate Transport across the Placenta
Folate (B9) is an essential molecule for the development and maturation of cells. During gestation, it is extremely important to the growth and development of the nervous system. Systemic deficiencies of B9 during pregnancy cause neural tube defects (NTD) ranging from spina bifida to anencephaly. However, over supplementation of folate has been linked with the progression of cancer and therefor...
متن کاملRegulation of Nutrient Transport across the Placenta
Abnormal fetal growth, both growth restriction and overgrowth, is associated with perinatal complications and an increased risk of metabolic and cardiovascular disease later in life. Fetal growth is dependent on nutrient availability, which in turn is related to the capacity of the placenta to transport these nutrients. The activity of a range of nutrient transporters has been reported to be de...
متن کاملCURRENT TOPIC Mineral transport across the placenta
We here review the transport of three minerals which have in common their role in bone mineralisation, their active transport across the placenta, and the rise in their placental transport rate in late gestation. The growing and differentiating human fetus is entirely dependent on its mother for the supply of nutrients and oxygen and removal of waste products. In mammals, through most of gestat...
متن کاملNotch1 controls development of the extravillous trophoblast lineage in the human placenta.
Development of the human placenta and its different epithelial trophoblasts is crucial for a successful pregnancy. Besides fusing into a multinuclear syncytium, the exchange surface between mother and fetus, progenitors develop into extravillous trophoblasts invading the maternal uterus and its spiral arteries. Migration into these vessels promotes remodelling and, as a consequence, adaption of...
